Reveals that mast cells release thymosin β4 under stress conditions, and that this Tβ4 contributes to intestinal epithelial barrier dysfunction in irritable bowel syndrome (IBS). In IBS patients, colonic mucosal Tβ4 was elevated. In Tβ4-deficient rats and mechanistic experiments, mast cell-derived Tβ4 impaired the IL22RA1/JAK1/STAT3 signaling pathway in epithelial cells. Demonstrates a paradoxical pro-pathological role for Tβ4 in a stress-IBS context—in contrast to its generally protective roles in other GI and injury models.
