Validates living myocardial slices (LMS) as an ex vivo model for testing cardiac regenerative gene therapies, including thymosin β4-based approaches. LMS from human and large-animal hearts maintain adult cardiomyocyte physiology for days. Tests confirmed that thymosin β4 gene delivery via viral vectors in LMS produced measurable improvements in contractile function and reduced injury markers. Establishes LMS as a translational platform between cell culture and in vivo models for evaluating TB4-based cardiac gene therapy.
