Tests thymosin β4 in DSS-induced colitis mice and finds that elevated Tβ4 in IBD paradoxically impairs intestinal mucus barrier integrity by inhibiting autophagy in goblet cells. Tβ4 overexpression reduced MUC2 secretion and mucus layer thickness through autophagy suppression. Conversely, autophagy activation reversed Tβ4-induced mucus barrier impairment. Identifies a pathological role for excess Tβ4 in IBD-associated intestinal barrier dysfunction, adding nuance to the generally protective role of Tβ4 in gastrointestinal contexts.
