Shows that endothelial Sirtuin1 (Sirt1) deficiency paradoxically improves skeletal muscle insulin sensitivity in mice through increased thymosin β4 secretion. Endothelial-specific Sirt1 knockout mice showed increased Tβ4 secretion into the circulation, which promoted insulin signaling in skeletal myotubes. Identifies an unexpected endothelium-to-muscle crosstalk axis mediated by Tβ4—relevant for understanding insulin resistance and potential metabolic applications of Tβ4.
