Tests thymosin β4 conditional knockout in hepatic stellate cells (HSCs) for liver fibrosis in a transgenic mouse model. Targeted Tβ4 deletion in HSCs (which normally activate and produce collagen in fibrosis) significantly reduced liver fibrosis—fibrosis score, collagen content, α-SMA expression, and Hedgehog pathway activation were all reduced compared to controls. Confirms Tβ4 expressed by HSCs is pro-fibrotic in the liver—a context where Tβ4 promotes rather than resolves fibrosis. Important contrast to Tβ4's anti-fibrotic roles in kidney and lung.
