Tests the relationship between p53 and PI3K/AKT signaling in medulloblastoma chemosensitivity, finding thymosin β4 as a key mediator. p53 upregulated TB4 expression, which promoted drug resistance through cytoskeletal remodeling and anti-apoptotic signaling. PI3K pathway inhibition combined with TB4 knockdown synergistically enhanced chemosensitivity. Confirms a p53-TB4-PI3K regulatory axis controlling medulloblastoma drug resistance and provides rationale for targeting TB4 to improve pediatric brain tumor treatment.
