Landmark proteomics discovery identifying thymosin β4 as a human exerkine (exercise-induced secreted factor) and growth factor. Contracting C2C12 myotubes secreted TB4 into media; plasma TB4 was acutely elevated in exercising humans and mice. Exerkine TB4 promoted cell proliferation, migration, and angiogenesis in recipient cells. Establishes skeletal muscle as a TB4 secretory organ activated by contraction—providing a new mechanistic link between exercise benefits (cardiovascular, tissue repair) and TB4 secretion.
