Identifies thymosin β4 as essential for platelet thrombus formation by controlling the G-actin/F-actin equilibrium. Tmsb4x-knockout mice showed severely impaired platelet activation, reduced thrombus formation in flow chambers, and prolonged bleeding times. Without TB4, platelets could not properly reorganize their actin cytoskeleton during activation, impairing filopodia/lamellipodia formation and clot retraction. Establishes TB4 as an endogenous platelet actin regulator critical for hemostasis—a pro-thrombotic role that counterbalances its anti-thrombotic vascular effects.
