Tests thymosin β4 overexpression for protection against oxygen-glucose deprivation/reperfusion (OGD/R) injury in rat cortical neurons—an in vitro model of ischemic stroke. TB4 overexpression significantly reduced OGD/R-induced neuronal apoptosis, preserved endoplasmic reticulum ultrastructure, decreased ER stress markers (GRP78, CHOP), and improved the Bcl-2/Bax survival ratio. Establishes ER stress suppression as a key mechanism for TB4's cortical neuroprotection—complementing its effects on mitochondrial preservation and providing a mechanistic target for TB4 in stroke and hypoxic brain injury.
