Tests whether thymosin β4 priming induces or expands Islet-1-positive cardiac progenitor cells in adult mice using a definitive genetic Isl1-nLacZ reporter line. Prior immunostaining-based studies claimed TB4 increased Islet-1 positivity, suggesting progenitor reactivation. In this rigorous genetic model, TB4 priming did not increase Islet-1-positive cell populations in adult ventricle. Provides an important negative result—challenging a proposed TB4 cardiac regeneration mechanism and highlighting the need for genetic reporter validation over immunostaining for rare progenitor cell populations.
