Uses high-speed atomic force microscopy (AFM) for live-cell imaging of cortical actin dynamics, directly measuring polymerization rates and filament synthesis frequency in living cells. Thymosin β4, as the primary G-actin sequestrant governing actin monomer availability for polymerization, provides conceptual context for the cortical actin dynamics observed. Advances fundamental understanding of how the TB4-managed G-actin pool feeds cortical filament networks—providing in vivo biophysical data complementing biochemical studies of TB4's actin sequestration function.
