KPV tripeptide was loaded into nanoparticles coated with hyaluronic acid, which targets both intestinal epithelial cells and macrophages — two key cell types involved in colitis. The nanoparticles were then embedded in a chitosan-alginate hydrogel to survive oral delivery and release in the colon. In a mouse colitis model, this system reduced intestinal damage and inflammation at far lower peptide doses than free KPV, with the hyaluronic acid coating proving critical for full therapeutic benefit.
