Demonstrates thymosin β4 acts as an intermediary between Rac1/Rap1 GTPase activation and cancer cell migration under hypoxia/reoxygenation (H/R) conditions. TB4 expression was required for H/R to activate these GTPases; inhibiting either TB4 or the GTPases reduced H/R-induced cell migration and metastasis. Establishes a TB4→Rac1/Rap1 GTPase axis mediating hypoxia-driven tumor invasion—mechanistically explaining how TB4 promotes cancer aggressiveness in the poorly oxygenated tumor microenvironment.
