Demonstrates that thymosin β4 knockdown in normal rat intestinal epithelial cells (IEC-6) triggers DNA re-replication—abnormal DNA amplification—by downregulating Emi1, an inhibitor of re-replication licensing. TB4 silencing caused cell cycle dysregulation in normal epithelial cells, not just cancer cells. Reveals a previously unknown role of TB4 in maintaining normal DNA replication fidelity in intestinal epithelia—adding a genome integrity function to TB4's repertoire and raising important considerations about unintended consequences of TB4 inhibition in CRC therapy.
