Dissects the interplay between thymosin β4, ADF/cofilin, and Arp2/3 complex in regulating F-actin treadmilling and HeLa cell migration. TB4 inhibited cofilin-stimulated actin treadmilling by sequestering monomers; active Arp2/3 complex reversed TB4's inhibition by providing branched nucleation sites. Immunofluorescence confirmed TB4 co-localization with cofilin and Arp2/3 in lamellipodia. Provides mechanistic resolution for how TB4, cofilin, and Arp2/3 compete and cooperate to control cell locomotion—relevant to understanding TB4's context-dependent pro- and anti-migration roles in wound healing versus cancer invasion.
