Develops a bacterial expression system for recombinant human thymosin β4 in protease-deficient E. coli BL21(DE3), testing three expression vectors to optimize yield. Soluble TB4 was purified by two-step immobilized metal ion affinity chromatography and confirmed active in endothelial cell angiogenesis assays. Provides an early-generation E. coli TB4 manufacturing approach—preceding the more advanced intein-based (PMID 27015974, 29989423) and high-density fermentation (PMID 23711379) systems—contributing to the evolving platform for cost-effective TB4 pharmaceutical production.
