Develops a high-yield E. coli production system for recombinant thymosin β4 using a DsbA-hexahistidine fusion tag in high cell density fed-batch fermentation (50 g/L dry cell weight). The fusion tag enabled soluble expression and efficient purification of biologically active TB4. Achieves pharmaceutical-scale TB4 production parameters—addressing a critical manufacturing bottleneck for TB4 clinical development and complementing the intein-based N-acetylation approaches (PMID 27015974, 29989423) with a high-density fermentation strategy for bulk TB4 production.
