Demonstrates that frog (Xenopus) limb regeneration capacity can be restored in post-metamorphic frogs—which normally cannot regenerate—by transplanting larval limb progenitor cells with activated Wnt/β-catenin signaling combined with Sonic hedgehog, FGF10, and thymosin β4. TB4 was required along with the other three factors for multidigit regenerate formation. Establishes TB4 as one of four critical signals for enabling limb regeneration in a gain-of-function model—providing fundamental evidence for TB4's role in limb progenitor cell activation and patterned tissue regeneration.
