This study examined intracellular signaling of alpha-MSH, KPV, and ACTH peptides in human keratinocytes. No cyclic AMP elevation was detected in response to these peptides in either HaCaT or normal keratinocytes. However, rapid intracellular calcium responses were observed in HaCaT cells treated with alpha-MSH and KPV at concentrations from femtomolar to nanomolar range, but only when the cAMP pathway was inhibited. Chinese hamster ovary cells transfected with MC-1R confirmed that alpha-MSH and KPV peptides can elevate intracellular calcium through this receptor, suggesting an alternative calcium-dependent signaling mechanism.
