This study demonstrates that obesity can be reversed without GLP-1 receptor activation. Researchers found that combined GIPR and GCGR agonism effectively reduces body weight in diet-induced obese mice. Retatrutide, a balanced GLP-1R/GIPR/GCGR triple agonist, normalized body weight even in GLP-1R knockout mice. A novel GIPR:GCGR co-agonist (BWB3054) with minimal GLP-1R activity achieved comparable weight loss. These findings suggest that the gastrointestinal side effects commonly associated with GLP-1 agonism might be avoidable while still achieving significant anti-obesity efficacy through GIP and glucagon receptor pathways.
