In vitro study in rotenone-treated primary retinal ganglion cells examining semaglutide's neuroprotective effects through improved mitochondrial function and glucose metabolism. Semaglutide protected retinal ganglion cells from metabolic and oxidative stress-induced degeneration via GLP-1R-mediated pathway activation. Provides mechanistic preclinical evidence for semaglutide as a neuroprotective agent in glaucoma—supporting epidemiological observations of reduced glaucoma incidence with GLP-1 RA therapy and motivating clinical trials of semaglutide for glaucoma neuroprotection.
