Generation and characterization of a humanized GLP-1 receptor (hGLP1R) mouse model by replacing rodent GLP1R with human GLP1R to enable preclinical pharmacodynamic testing of non-peptide oral small-molecule GLP-1 RAs that are inactive at the rodent receptor. Validates the model using established GLP-1 RA pharmacodynamic endpoints. Addresses a critical translational gap for small-molecule GLP-1 RA development—enabling preclinical efficacy and dose-finding studies for oral non-peptide GLP-1 RAs (such as orforglipron) that cannot be tested in standard rodent models.
