Review of MOTS-c's protective roles in acute lung injury—covering its anti-inflammatory, antioxidant, anti-ferroptotic, and endothelial barrier-preserving mechanisms across different ALI models including LPS-induced, ischemia-reperfusion, sepsis-associated, and radiation-induced lung injury. Synthesizes the growing ALI-focused MOTS-c evidence. Establishes MOTS-c as a multi-mechanism lung protective agent for ALI—a syndrome with high mortality and no approved pharmacological therapy—providing a rationale for clinical investigation of MOTS-c as a treatment for ARDS across diverse etiologies.
