This study demonstrated that the natural inhibition of growth hormone secretion during trough periods between pulses is driven by somatostatin release triggered by the preceding growth hormone peak. When endogenous GHRH was blocked by antibodies, the periodic suppression of growth hormone disappeared, but it could be restored by infusing exogenous growth hormone to mimic natural pulses. Blocking somatostatin completely prevented this growth hormone-induced suppression, confirming that the pulsatile pattern of growth hormone secretion relies on a somatostatin-mediated feedback loop.
