Six short children with low growth hormone secretion received continuous subcutaneous GHRH infusions for three weeks, and pulsatile growth hormone release was restored in most of those given the higher dose. However, the magnitude of the growth hormone response was generally lower on day 21 compared to day 1, suggesting some degree of desensitization over time. Blood levels of GHRH were higher on day 21 than day 1, indicating changed drug metabolism, and the researchers suggest that a long-acting GHRH analogue may be needed for optimal long-term treatment.
