This study tracked when growth hormone abnormalities emerge in genetically obese Zucker rats and found that the defect appears by eight weeks of age. At six weeks, obese rats still responded normally to growth hormone-releasing factor, but by eight weeks their response was significantly blunted both in live animals and in isolated pituitary cells. The impairment appeared to involve both central factors like increased hypothalamic somatostatin and peripheral factors, since serum from obese rats could suppress growth hormone release from normal pituitary cells.
