This study revealed that the tumor suppressor protein p21 is essential for the anti-cancer effects of GHRH antagonists. When p21 was knocked down in lung cancer, colon cancer, and bone cells, the GHRH antagonist JMR-132 lost its ability to inhibit cell growth, showing that GHRH antagonists work through a p21-dependent mechanism.
