NMR analysis of bovine growth hormone-releasing factor analogs revealed that while truncating the first two amino acids drastically reduced biological potency (by 10,000-fold), the overall helical structure in the 8-29 region remained virtually identical to more active forms. This disconnect between structure in solution and biological activity suggests that the N-terminal residues are critical contact points for receptor binding rather than being important for the peptide's three-dimensional shape.
