This study investigated thymosin alpha-1's effects on pituitary hormone release in rats. When injected into the third ventricle, it significantly decreased plasma levels of TSH, ACTH, and prolactin but did not affect growth hormone. Interestingly, when hemipituitaries were incubated directly with the peptide in vitro, thymosin alpha-1 stimulated TSH and ACTH release in a dose-dependent manner while not affecting prolactin or growth hormone. This suggests thymosin alpha-1 has opposing effects on pituitary function depending on the route—inhibitory centrally (via hypothalamus) but stimulatory at the pituitary level.
