This study investigated how hexarelin, a synthetic growth hormone-releasing peptide, stimulates GH secretion from human pituitary tumor cells. Hexarelin worked through a protein kinase C-dependent mechanism that is distinct from the pathway used by GHRH, and a GHRH receptor blocker could not prevent hexarelin's effects. Interestingly, when combined with GHRH, hexarelin amplified the cAMP signaling response, suggesting these two GH-releasing pathways can work together synergistically.
