The GHRH receptor rapidly becomes less responsive after brief exposure to GHRH, a process called desensitization, as demonstrated in both engineered cell lines and rat pituitary cells. In pituitary cells, the growth hormone release response was more sensitive to this desensitization than the cAMP signaling response, and the receptor was internalized from the cell surface after GHRH exposure. This desensitization mechanism has implications for understanding why repeated or continuous GHRH stimulation may become less effective over time.
