While GHRH gene transcripts were found in 5 of 9 human pituitary growth hormone-secreting tumors, a GHRH antagonist failed to inhibit their baseline growth hormone secretion or cAMP production. The antagonist effectively blocked the effects of externally added GHRH, confirming it was functional, but the lack of baseline inhibition argues against the theory that tumor-produced GHRH creates a self-stimulating growth loop in these pituitary tumors.
