Using a competitive GHRH receptor antagonist as a measuring tool, researchers demonstrated that elderly men produce less hypothalamic GHRH than young men, contributing to their lower growth hormone levels. The antagonist suppressed spontaneous GH secretion more easily in elderly subjects, indicating less endogenous GHRH was competing for the receptor. This provides the first in-vivo evidence that reduced GHRH output from the brain is a key driver of age-related growth hormone decline.
